CYTOTOXIC ACTIVITY TEST OF p-DIMETHYLAMINOCHALCONE COMPOUNDS AND CHALCONE AGAINTS MCF-7 AND VERO CELL


ABSTRACT

One common type of cancer that affects women is breast cancer. Various therapies have not been able to cure breast cancer. Chalcone compounds have promising anticancer activity. ρ-dimethylaminochalcone compound is a derivative of the chalcone. This research was conducted to determine cytotoxic activity of ρ-dimethylaminochalcone compared with chalcone against the MCF-7 and Vero cells.
MTT Test was used as the method in the cytotoxic activity test in the MCF-7 and Vero cells. The series of concentration used in the MCF-7 cell for the ρ-dimethylaminochalcone and chalcone were 100; 50; 25; 12,5; 6,25; 3,12; 1,56 μg/mL. In the Vero cell, the series of concentration used for ρ-dimethylaminochalcone and chalcone were 32; 16; 8; 4; 2; 1; 0,5 μg/mL. The Absorbance of the cell was measured at 550 nm wavelength using the ELISA reader. IC50 was used as a cytotoxicity parameter that was obtained with probit analysis.
The value of IC50 to the MCF-7 cell from the ρ-dimethylaminochalcone was 4,49 µg/mL and 1,36 µg/mL for chalcone. The value of IC50 to the MCF-7 cell from ρ-dimethylaminochalcone was 47,59 µg/mL dan 13,98 µg/mL for chalcone. It shown that the ρ-dimethylaminochalcone had a lower cytotoxic activity than chalcone to MCF-7 and Vero cell.
